Vitamin D inhibition of the prostaglandin pathway as therapy for prostate cancer.

چکیده
Prostaglandins have been shown to play a role in carcinogenesis and the progression of many cancers, including prostate cancer. We have recently discovered that the biologically active form of vitamin D, 1,25(OH)2D or calcitriol, regulates multiple genes involved in the prostaglandin pathway. These effects include decreasing prostaglandin synthesis, increasing prostaglandin catabolism, and inhibiting the expression of prostaglandin receptors. In combination, these three mechanisms reduce prostaglandin levels and signaling, thereby attenuating the growth-stimulatory effects of prostaglandins in prostate cancer.

Imaging Cellular Proliferation in Prostate Cancer with Positron Emission Tomography
Prostate cancer remains a major public health problem worldwide. Imaging plays an important role in the assessment of disease at all its clinical phases, including staging, restaging after definitive therapy, evaluation of therapy response, and prognostication. Positron emission tomography with a number of biologically targeted radiotracers has been demonstrated to have potential diagnostic and...
متن کاملMechanisms of vitamin D-mediated growth inhibition in prostate cancer cells: inhibition of the prostaglandin pathway.
Calcitriol (1,25-dihydroxyvitamin D3), the active form of vitamin D, promotes growth inhibition and differentiation in prostate cancer (PCa) cells. To unravel the molecular pathways of calcitriol actions, cDNA microarray analysis was used to identify novel calcitriol target genes including two that play key roles in the metabolism of prostaglandins (PGs), known stimulators of PCa growth and pro...
متن کاملQSAR Study of 17β-HSD3 Inhibitors by Genetic Algorithm-Support Vector Machine as a Target Receptor for the Treatment of Prostate Cancer
The 17β-HSD3 enzyme plays a key role in treatment of prostate cancer and small inhibitorscan be used to efficiently target it. In the present study, the multiple linear regression (MLR),and support vector machine (SVM) methods were used to interpret the chemical structuralfunctionality against the inhibition activity of some 17β-HSD3inhibitors. Chemical structuralinformation were described thro...

متن کاملThe Role of Adiponectin in Prostate Cancer: A Narrative Review
Prostate cancer (PCa) is the most common type of cancer among men over 60 years old. The aggressiveness and mortality of PCa can be correlated with obesity. Adipose tissue-derived cytokines such as adiponectin may explain the correlation between PCa and obesity. Since the correlation between adiponectin and aggressive PCa is still not fully evaluated, we aimed to investigate the probable role o...
